Tulisokibart  (Synonyms: PRA023)

Tulisokibart (PRA023) is a humanized IgG1-κ monoclonal antibody. Tulisokibart targets to TNFSF15/TL1A. Tulisokibart can be used to study a variety of inflammatory/fibrotic diseases, such as Crohn's Disease (CD) and ulcerative colitis^[1][2].

Tulisokibart successfully reduces body weight loss, disease active index (DAI) scores and inflammatory cytokines release in humanised TL1A mice. Histological analysis shows reverse inflammatory cell infiltration, crypt structure change and fibrosis in humanised mouse models for inflammatory bowel disease^[3].

  Human IgG1 kappa

• 
  Flow cytometric analysis of 1×10⁶ MCF-7 cells labelling TL1A (red) with Tulisokibart (HY-P990007). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

  Immobilized TL1A/TNFSF15 Protein, Human can bind Tulisokibart. The EC₅₀ for this effect is 15.31 ng/mL.

  Loaded Tulisokibart on AHC2 biosensor, can bind TL1A/TNFSF15 Protein, Human (HY-P71913) with an affinity constant of 3.693E-10 M as determined in BLI assay.
