TNFRSF8/CD30

TNFRSF8/CD30 is a TNF receptor superfamily member expressed on activated lymphocytes and characteristically detected in Hodgkin lymphoma and anaplastic large-cell lymphoma[1]. Mechanistically, CD30 recruits TRAF2 and TRAF5 to activate NF-κB, and studies in anaplastic large-cell lymphoma cells show activation of both canonical and alternative NF-κB pathways[2][3]. In disease models, CD30 ligand and agonistic anti-CD30 antibodies produce pleiotropic effects in CD30-positive lymphoma cell lines, including reduced proliferation and cytolytic cell death in large-cell anaplastic lymphoma cells[4]. In Hodgkin lymphoma cell lines, targeted repression of overexpressed CD30 downregulates NF-κB and ERK1/2, supporting CD30 as a functional signaling node rather than only a diagnostic marker[5]. Compared with membrane CD30, soluble CD30 binds CD153/CD30L with high affinity and blocks transmembrane CD30 signaling, making isoform context important for experimental design[6]. For applications, anti-CD30 antibody-drug conjugate cAC10-vcMMAE shows potent and selective activity against CD30-positive tumor lines and xenograft models[7].
