TNFRSF4

TNFRSF4 (OX40/CD134) is an inducible T-cell costimulatory receptor that signals through TRAF2/TRAF5-mediated NF-κB activation[1]. Mechanistically, OX40 supports antigen-driven CD4+ T-cell persistence by maintaining BCL2 and BCL2L1/BCL-xL expression, while sustained survivin expression drives clonal expansion[2][3]. In disease models, the OX40L-OX40 axis promotes aberrant T follicular helper responses in systemic lupus erythematosus and marks immune escape by Tnfrsf4-expressing regulatory T cells in chronic myeloid leukemia[4][5]. Compared with membrane OX40, a soluble OX40 isoform has been identified in serum from healthy donors and patients with autoimmune disease or cancer, supporting isoform-aware assay design[6]. For experimental applications, anti-OX40 agonist antibodies such as MOXR0916 have been evaluated in advanced solid tumors to test effector T-cell costimulation and regulatory T-cell modulation[7].
