Description

H-Trp-Phe-OH (Trp-Phe) is an ACE inhibitor and endothelial function regulator, with an ACE IC50 of 0.318 mg/mL. H-Trp-Phe-OH increases nitric oxide concentration, reduces endothelin-1 (ET-1) levels, and reverses norepinephrine-mediated endothelial cell dysfunction. H-Trp-Phe-OH exhibits antihypertensive activity. H-Trp-Phe-OH can be used in studies related to hypertension and atherosclerosis[1].

In Vitro

H-Trp-Phe-OH (Trp-Phe) (200-300 μM; 24 h) does not cause significant cytotoxicity in HUVECs[1].
H-Trp-Phe-OH (Trp-Phe) (50-200 μM; 24 h) increases NO production in HUVECs in a concentration-dependent manner (up to 2.35 μmol/gprot at 200 μM over 24 h) and reverses norepinephrine-induced reductions in NO production in HUVECs[1].
H-Trp-Phe-OH (Trp-Phe) (50-200 μM; 24 h) decreases ET-1 production in HUVECs and reverses norepinephrine-induced increases in ET-1 production in HUVECs, with significant activity observed at 50, 100, and 200 μM over 24 h[1].

ELISA Assay[1]

Cell Line: human umbilical vein endothelial cells (HUVECs)
Concentration: 50 μM; 100 μM; 200 μM; 200 μM (in the presence of 0.5 μM norepinephrine)
Incubation Time: 24 h
Result: Significantly decreased ET-1 production relative to blank control HUVECs.
Significantly reduced the norepinephrine-induced increase in ET-1 production in HUVECs, with the effect of 200 μM Trp-Phe being greater than that of other tested peptides.
