d-KLA Peptide (D-(KLAKLAK)2), KLA peptide (HY-P5345) isomer, is a pro-apoptosis peptide. d-KLA Peptide can specifically target mitochondria and induce apoptosis by destroying the mitochondrial membrane. d-KLA Peptide increases caspase 3/7 activity, exerts proapoptotic activity and enhances antitumor efficacy in mouse melanoma models[1].

d-KLA Peptide (400 μM) kills 50% of eukaryotic cells in the monolayer[1].
d-KLA Peptide induces mitochondrial swelling in a cell-free system with an ED50 of 0.44 μM[1].
d-KLA Peptide induces mitochondrial membrane potential loss in a cell-free system with an ED50 of 0.4 μM[1].
d-KLA Peptide (1.5 μM in 1 μM ODN; 24 h) can form liposomes, which can reduce the viability of B16 (F10) mouse melanoma cells and enhance caspase 3/7 activity[1].

In Vivo

Cationic liposome-loaded d-KLA Peptide (1.52 µg)/G3139 (4 µg) (intratumoral; twice daily for 2 doses) provides significant antitumor efficacy in melanoma-bearing mice, achieving 60% tumor growth inhibition via enhanced apoptotic activity[1].

Animal Model: C57BL/6 (male, initial weight 24.4 g, subcutaneous B16(F10) melanoma model)[1]
Dosage: 1.52 µg with G3139 (4 µg) in cationic liposome
