Background

Contactin-1 (CNTN1) protein, a key player in nervous system development, facilitates critical cell surface interactions. It plays a significant role in the formation of paranodal axo-glial junctions within myelinated peripheral nerves and contributes to signaling between axons and myelinating glial cells by associating with CNTNAP1. Additionally, CNTN1 participates in the generation of oligodendrocytes, acting as a ligand for NOTCH1. The association with NOTCH1 promotes the activation of NOTCH1, leading to the release of the notch intracellular domain (NICD) and its subsequent translocation to the nucleus. Notably, CNTN1's interaction with TNR induces neuron repulsion and inhibits neurite outgrowth. In its monomeric form, CNTN1 interacts with CNTNAP1 in cis, and it binds to the carbonic-anhydrase like domain of PTPRZ1. Furthermore, CNTN1 forms a complex with NRCAM and PTPRB and interacts with TASOR. These multifaceted interactions underscore the diverse functions of Contactin-1 in orchestrating crucial processes during nervous system development.
