3BP-3940 (50 μg; for Ga-68 radiolabeling): During the automated radiolabeling process of [Ga]-3BP-3940, reacting at 85°C in a labeling volume of 7mL with 0.7M, pH5.5 sodium acetate buffer (2 mL) for 20 minutes, decay-corrected radiochemical yield (RCY) ≥90% was achieved. The final product activity after automated synthesis ranged between 308 MBq-987 MBq (decay-corrected RCY=93.1 %). Radiochemical purity (RCP) was validated to be ≥97%, with an average RCP of 97.7±0.9% in subsequent routine production[1].
3BP-3940 (37-50 mg/GBq; for Lu-177 radiolabeling): In the automated radiolabeling process of [Lu]-3BP-3940, high RCY results were obtained by reacting with a 0.1M ascorbic acid solution containing gentisic acid (1.2 mL, pH4.5, 30mg gentisic acid) at 85°C for 30 minutes. The average RCP from process validation runs was 97.5±1.4%, with an average yield of 94.4±1.6%. In subsequent routine production, the average RCP at the end of synthesis was 96.8±1.2%, and the average RCY was 91.5±9.1%[1].
[Ga]-3BP-3940 and [Lu]-3BP-3940 when incubated in human serum (HS) and saline (NaCl) at 37°C showed no significant release of radioactive metals over two half-lives of their respective isotopes, demonstrating complex stability >90% under both conditions. Both compounds had an RCP > 95% as measured by HPLC within 2 hours post-formulation[1].
3BP-3940 (concentration for UV detection: 0.5-10 mg/mL, concentration for radioactivity detection: 0.1-1.0 MBq; HPLC method validation): In the HPLC method validation of [Ga]-3BP-3940, linearity, accuracy, precision, specificity, lower limit of detection (LLOD), and lower limit of quantification (LLOQ) all met the requirements of ICH guideline Q2 and EANM guidelines for radiopharmaceuticals validation[1].
