MCI-INI-3 (0-100 μM; 30 min) potently and selectively inhibits recombinant human ALDH1A3 (IC50 = 0.46 μM; Ki = 0.55 μM) with ~140-fold greater selectivity over recombinant human ALDH1A1 (Ki = 78.2 μM) via competitive inhibition of the aldehyde substrate[1].
MCI-INI-3 (0.015-15 μM; 15 min-6 day) potently inhibits ALDH activity in U87MG glioblastoma cells and GSC-326 mesenchymal glioma stem cells, with sustained inhibition lasting up to 6 days in GSC-326 cells[1].
MCI-INI-3 (15 μM; 5 day) reduces proliferation of both MES (GSC-83, GSC-326) and PN (GSC-19, GSC-84) glioma stem cells at 15 μM, but this effect is not ALDH1A3-specific, as shown by similar IC50 values in GSC-326 wild-type and ALDH1A3-knockout cells[1].
MCI-INI-3 (15 µM) reduces RA production in U87MG glioma stem cells at 15 µM but shows non-specific cytotoxicity against non-ALDH1A3-expressing glioma stem cell lines at the same concentration[2].
