LIN28-IN-2 is a Lin28 inhibitor with activity against Lin28a, Lin28b, and their zinc knuckle domain. LIN28-IN-2 blocks Lin28-RNA substrate binding, perturbs zinc knuckle domain conformation. LIN28-IN-2 inhibits cancer cell proliferation, spheroid growth, and induces G2/M phase arrest. LIN28-IN-2 suppresses cancer stem cell phenotypes, Lin28-mediated stress granule formation, let-7 target genes, cancer stem cell biomarkers, and neuroendocrine biomarkers expression in cancer cells. LIN28-IN-2 can be used for the research of cancer.

LIN28-IN-2 (Compound Ln268) (0-500 μM) dose-dependently inhibits the interaction between purified Lin28b ZKD protein and pre-let-7d RNA in an EMSA biochemical assay[1].
LIN28-IN-2 (20 μM; 72 h) suppresses Lin28b activity in DuNE cells by increasing let-7d levels, reducing Lin28b expression, and downregulating downstream let-7 targets, CSC biomarkers, and NE biomarkers[1].
LIN28-IN-2 potently inhibits proliferation of Lin28b-positive DuNE cells and Lin28a-positive IGROV-1 cells, with stronger activity than parent compound Ln15[1].
LIN28-IN-2 (0-100 μM; 0-6 days) inhibits 3D spheroid growth of Lin28b-positive DuNE cells with an IC50 of 10 μM[1].
LIN28-IN-2 (14 days) strongly inhibits colony formation of Lin28b-positive DuNE cells, reducing colony counts by 96% after 14 days of treatment[1].
LIN28-IN-2 (0-20 μM; 144 h) induces dose-dependent G2/M phase cell cycle arrest in Lin28b-positive DuNE cells[1].
LIN28-IN-2 selectively inhibits proliferation of Lin28-positive cancer cell lines (including DuNE, IGROV-1, TOV-112D, Kuramochi, AN3CA, and VOA1066) and has no effect on Lin28-negative cell lines[1].
LIN28-IN-2 inhibits Lin28b-mediated stress granule formation in DuNE cells by reducing Lin28b SG localization and blocking SG maturation[1].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (22.10 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 5 mg/mL (22.10 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
