This genomic element is located approximately 32 kb from the 3' end of the Shox2 (short stature homeobox 2) gene. This region functions as an enhancer necessary for expression of the Shox2 gene in mouse embryo limb and hindbrain. It is conserved in vertebrates and includes two sub-regions with deeper conservation, LHB-A and LHB-B, which drive more specific expression patterns in limb and neural tissue, respectively. An ultraconserved region of LHB-B is also homologous to part of the conserved non-coding element 5 (CNE5) neural enhancer found downstream of the homologous human SHOX (short stature homeobox) gene in the pseudoautosomal region of chromosomes X and Y, though mouse lacks an equivalent paralogous gene, and it thus represents a duplicated conserved non-coding element (dCNE).
