Pregnane X receptor (or NR subfamily 1, group I, member 2, NR1I2), a ligand-activated nuclear receptor (NR), regulates the expression of metabolic enzymes and transporters involved in the response of mammals to their chemical environment. PXR has a wide spectrum of ligands, including prescription drugs, herbal medicines, dietary supplements, environmental pollutants, and endobiotics. Upon ligand binding, PXR forms of a heterodimer with retinoid X receptor (RXR), and subsequently binds to PXR response elements in the 5′-flanking region of the PXR target genes, resulting in the transcriptional activation. Among the target genes are those encoding including both phase I and phase II metabolic enzymes, as well as transporters which contribute to absorption, distribution, metabolism and elimination of xenobiotics and endobiotics.
