How CETSA Works
CETSA is based on the principle that drug binding can alter the thermal stability of target proteins. Proteins are structurally stable under physiological conditions, however, as temperature rises, they gradually denature, transforming from a soluble state to an insoluble precipitate. For certain proteins, the melting temperature (Tm) is defined as the temperature at which 50% of the protein denatures. The Tm value reflects the stability of the protein structure. When small molecules (such as drugs) bind to target proteins, they increase the thermal stability of the target proteins. As the temperature increases, unbound proteins denature first, while drug-bound proteins remain relatively stable. Using techniques such as western blot or mass spectrometry (MS), the extent of protein degradation at different temperatures can be detected, allowing for the inference of drug binding to the target protein.
