CBFA2T3 is a transcriptional corepressor that facilitates gene silencing by associating with DNA-binding transcription factors and recruiting other corepressors or histone-modifying enzymes (e.g., EGLN1). It represses MMP7 expression in a ZBTB33-dependent manner and promotes HIF1A prolyl hydroxylation-dependent ubiquitination and degradation via interaction with EGLN1, thereby reducing HIF1A protein stability. Additionally, CBFA2T3 downregulates glycolytic genes (e.g., PFKFB3, PFKFB4, PDK1, PFKP, LDHA, HK1)—direct targets of HIF1A—to suppress glycolysis and enhance mitochondrial respiration. It regulates erythroid progenitor proliferation/differentiation by inhibiting TAL1 target genes (by similar) and contributes to granulocyte differentiation.
