UBCH7 is a ubiquitin-conjugating enzyme E2 that specifically functions with HECT-type and RBR family E3 ubiquitin-protein ligases. Unlike most RING-containing E3 ligases, it lacks intrinsic E3-independent reactivity with lysine but exhibits activity with RBR family E3 enzymes (e.g., PRKN, RNF31, ARIH1), which operate like RING-HECT hybrids. It accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to target proteins. By similarity, it mediates ubiquitination by the CUL9-RBX1 complex. In vitro, it catalyzes 'Lys-11'-linked polyubiquitination and participates in the selective degradation of short-lived or abnormal proteins. Downregulated during the S-phase, it contributes to cell cycle progression. Additionally, it regulates nuclear hormone receptor transcriptional activity and may play a role in myelopoiesis.
