  • Function

    HPV18 E6 is the core oncoprotein encoded by high-risk human papillomavirus type 18, approximately 150-160 aa. It contains two zinc finger domains and a C-terminal PDZ binding motif. It has no enzymatic activity itself and exerts its carcinogenic effect by hijacking host proteins. Its core function is to form a heterodimer with the E3 ubiquitin ligase E6AP, recruiting and polyubiquitinizing p53, causing it to be degraded by the proteasome, thereby abolishing its "genome guardian" function. In addition, E6 synergistically inhibits p53 through multiple mechanisms, including binding to CBP/p300, inhibiting p53 phosphorylation, and cytoplasmic isolation; it degrades PDZ domain tumor suppressor proteins such as DLG1, SCRIB, and MAGI via C-terminal PBM, disrupting cell polarity and tight junctions; it independently activates hTERT/telomerase to achieve cell immortalization; it degrades Bak to inhibit apoptosis; and it binds to IRF3 to achieve immune escape. E6 is a key driver and important target of cervical cancer (especially HPV18-related cervical adenocarcinoma, such as HeLa cells) and other HPV-related malignancies.
