COX-2-IN-69

COX-2-IN-69 is a selective, orally active COX-2 inhibitor with an IC₅₀ of 0.90 μM. COX-2-IN-69 occupies the secondary pocket of COX-2, interacts with His90 and Arg513, and reduces serum levels of PGE₂, IL-1β and TNF-α. COX-2-IN-69 exerts anti-inflammatory activity in a rat paw swelling model. COX-2-IN-69 can be used in inflammation-related research.

COX-2-IN-69 (Compound 4c) (10 mg/kg; p.o.; single administration) exhibits potent in vivo anti-inflammatory activity in a rat paw edema model^[1].

Sprague Dawley (male, 130-150 g, Carrageenan (HY-125474)-induced paw edema model)^[1]

Result:

Reduced paw edema percentage to 9.91% at 3 hours post-Carrageenan injection.
Showed no significant difference in paw volume compared to normal control at 2 and 3 hours.
Reduced serum PGE2 levels by 59.37%, serum IL-1β levels by 57.81%, and serum TNF-α levels by 54.70% compared to the Carrageenan control group.
Caused moderate desquamation of gastric mucosa, inflammatory cell infiltration of the submucosa layer, and vacuolar degeneration of gastric glands.
