HPPD-IN-10
HPPD-IN-10 is an inhibitor of Arabidopsis thaliana 4-hydroxyphenylpyruvate dioxygenase (HPPD), with an IC50 of 0.06 μM and a Kd of 0.265 μM. HPPD-IN-10 blocks the HPPD-mediated catalytic conversion of 4-hydroxyphenylpyruvate, downregulates the biosynthesis of plastoquinone and tocopherol, induces weed bleaching, and inhibits root and shoot growth. HPPD-IN-10 can be used in herbicide-related research on gramineous and broadleaf weeds in farmlands, such as barnyard grass and purslane.

HPPD-IN-10 (compound A3) potently inhibits Arabidopsis thaliana 4-hydroxyphenylpyruvate dioxygenase (HPPD), with an IC50 value of 0.06 μM[1].
HPPD-IN-10 (10-100 μg/mL; 5-day incubation) exhibits strong pre-emergence herbicidal activity against germinated Portulaca oleracea seeds and moderate activity against germinated Echinochloa crusgalli seeds; at the concentration of 100 μg/mL, its root inhibition rate on Portulaca oleracea reaches 83.4% and shoot inhibition rate reaches 92.2%[1].
HPPD-IN-10 binds to AtHPPD through bidentate chelation with Co2+, π−π interactions with Phe381 and Phe424, hydrophobic interactions with Gln293 and Leu265, and hydrogen bonding with Asn282[1].
HPPD-IN-10 binds tightly to wild-type Arabidopsis thaliana HPPD with a Kd value of 0.265 μM; however, its binding affinity to the AtHPPDN282A mutant is significantly reduced, which confirms that Asn282 is a key binding site[1].
