Oxonol VI is an optical indicator of membrane potential in lipid vesicles (excitation/emission wavelengths: 614/646 nm). Oxonol VI can be used to detect changes in membrane potential associated with (Na+ + K+)-ATPase activity in reconstituted vesicles.

Guide (The following is our recommended protocol. This protocol is for guidance only and should be modified according to your specific needs).
1 Preparation of dye stock solution
1) Prepare a 3.16 mM Oxonol VI stock solution in ethanol.
2 Preparation of dye working solution
1) Dilute the 3.16 mM Oxonol VI stock solution with a mixture of ethanol and water at a volume ratio of 1:5 (ethanol:water).
2) Adjust the dilution ratio to ensure that the final concentration of Oxonol VI reaches the range of 10-500 nM.
3. Specific steps
1) Add 1 mL of the appropriate buffer to the fluorescence cuvette and equilibrate to the set temperature (20 °C by default unless otherwise specified).
2) Measure the background fluorescence intensity of the buffer in the cuvette.
3) Add 5 μL of the prepared Oxonol VI working solution to the cuvette containing the equilibrated buffer.
4) After the fluorescence signal stabilizes, add a predetermined amount of vesicle suspension to the cuvette.
5) Continuously monitor the fluorescence signal using a spectrophotometer. Calculate the relative fluorescence change caused by the addition of the vesicle suspension, using the baseline fluorescence intensity recorded before the addition of the vesicles as a reference.
