DOPE-PEG5000-GE11

DOPE-PEG5000-GE11 is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against EGFR (HER1) on the surface of various epithelial tumor cells (lung cancer, breast cancer, colorectal cancer, head and neck cancer). Composition and role of each component:
1. PEG5000 (polyethylene glycol, MW 5000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. GE11 (GE11 peptide (YHWYGYTPQNVI)): Targeting ligand-a 12-amino acid peptide identified through phage display screening that selectively binds to epidermal growth factor receptor (EGFR/HER1) without activating its downstream signaling cascade. Enables receptor-mediated endocytosis into EGFR-overexpressing tumor cells while avoiding pro-proliferative signaling, making it a safer alternative to EGF for targeted delivery.
