EHMT1 is a histone methyltransferase that specifically mono- and dimethylates lysine 9 of histone H3 (H3K9me1 and H3K9me2) in euchromatin. H3K9me serves as an epigenetic mark for transcriptional repression by recruiting HP1 proteins to methylated histones. It also weakly methylates lysine 27 of histone H3 (H3K27me) and is involved in DNA methylation (with its histone methyltransferase activity functioning independently of DNA methylation). Likely targeted to histone H3 by DNA-binding proteins such as E2F6, MGA, MAX, or DP1, it contributes to silencing MYC- and E2F-responsive genes during G0 phase, implicating a role in G0/G1 transition regulation. Beyond histone methylation, EHMT1 dimethylates non-histone proteins (e.g., lysine 373 of p53/TP53) and represses mitochondrial function-related genes, potentially through promoter occupancy in concert with BAZ2B (by similarity).
