LAMTOR2 Protein, Human (His)

Cat. No.: HY-P77053
Handling Instructions

LAMTOR2, a pivotal component of the Ragulator complex, centralizes amino acid sensing and mTORC1 activation, responding to stimuli like growth factors, energy levels, and amino acids. As an amino acid-activated guanine nucleotide exchange factor (GEF), the Ragulator complex recruits Rag GTPases (RagA/RRAGA, RagB/RRAGB, RagC/RRAGC, and/or RagD/RRAGD) to lysosome membranes. This orchestrated process acts as a scaffold, guiding mTORC1 to lysosomes for activation. LAMTOR2 additionally acts as an adapter protein, enhancing the MAP kinase cascade's efficiency, facilitating MAPK2 activation. Interacting with LAMTOR1 and LAMTOR3, LAMTOR2 is integral to the lysosomal folliculin complex (LFC) and Ragulator complex, crucial for cellular processes. LAMTOR2 Protein, Human (His) is the recombinant human-derived LAMTOR2 protein, expressed by E. coli, with N-His labeled tag. The total length of LAMTOR2 Protein, Human (His) is 125 a.a., with molecular weight of ~13 kDa.

Background

As a crucial component of the Ragulator complex, LAMTOR2 plays a central role in amino acid sensing and the activation of mTORC1, a signaling complex that promotes cell growth in response to various stimuli, including growth factors, energy levels, and amino acids. Activated by amino acids, the Ragulator complex functions as a guanine nucleotide exchange factor (GEF) for the small GTPases Rag (RagA/RRAGA, RagB/RRAGB, RagC/RRAGC, and/or RagD/RRAGD), simultaneously mediating the recruitment of Rag GTPases to the lysosome membrane. This orchestrated activation of the Ragulator and Rag GTPases serves as a scaffold, recruiting mTORC1 to lysosomes, where it is subsequently activated. Additionally, LAMTOR2 acts as an adapter protein enhancing the efficiency of the MAP kinase cascade, facilitating the activation of MAPK2 (By similarity). LAMTOR2 is part of the Ragulator complex, interacting with LAMTOR1 and LAMTOR3, and is a key player in the lysosomal folliculin complex (LFC), which includes FLCN, FNIP1 (or FNIP2), RagA/RRAGA or RagB/RRAGB GDP-bound, RagC/RRAGC or RagD/RRAGD GTP-bound, and Ragulator.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
