NR162 is a selective CASK (Ca2+/calmodulin-dependent Ser/Thr kinase) inhibitor with an IC50 of 80 nM and a Kd of 22 nM. NR162 shows about 50-fold selectivity for CASK than TYRO3. NR162 targets the unique GFG motif of CASK and has excellent shape complementarity to the CASK ATP binding pocket. NR162 can be used for the research of neurological diseases.

NR162 (1 μM) shows excellent selectivity for CASK in kinome-wide profiling. NR162 weakly inhibits TYRO3 and CSNK1E, with IC50 values of 3.79 μM and 10.9 μM, respectively, and shows no inhibitory effect on MERTK, AXL, and ABL1[1].
NR162 (1 μM; 6 days) does not impair differentiation or affect β3-tubulin abundance in P19 cells[1].
NR162 (2 μM) causes cell death in differentiating P19 cells[1].
NR162 (10 μM) has no effect on growth or toxicity in NCI60 cancer cell lines[1].
