2-Carboxybenzaldehyde (Phthalaldehydic acid) is a key intermediate metabolite in the biodegradation process of polycyclic aromatic hydrocarbons, and also a specific substrate for CBA reductase and NAD-dependent 2-carboxybenzaldehyde dehydrogenase. 2-Carboxybenzaldehyde undergoes regioselective reduction via CBA reductase in mammalian tissues to produce 2-hydroxymethylbenzoic acid. 2-Carboxybenzaldehyde is converted to phthalic acid by NAD-dependent dehydrogenase during bacterial degradation. 2-Carboxybenzaldehyde can be used in relevant research in fields such as organic synthesis.

2-Carboxybenzaldehyde (CAB) (1 mM; 10 min at 37°C) is the preferred substrate of CBA reductase purified from the liver cytosol of phenobarbital-treated rats. It is converted to 2-hydroxymethylbenzoic acid in an NADH-dependent manner under the catalysis of CBA reductase. The apparent Km and Vmax values of this enzyme for 2-Carboxybenzaldehyde are 1.7 mM and 27.6 μmol/min per mg protein, respectively[1].
2-Carboxybenzaldehyde is specifically oxidized to phthalic acid by the purified NAD-linked dehydrogenase derived from Alcaligenes faecalis strain AFK2, with a Km value of 54 μM[2].
2-Carboxybenzaldehyde coordinates with Ca2+ ions to form an eight-coordinate monoclinic (space group C2/c) one-dimensional chain-like Ca2+ coordination polymer[3].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
