Reozalimab

Reozalimab (LY3434172; IBI-318) is an IgG1 monoclonal antibody targeting PD-1/PD-L1, with an IC50 of 2.27 nM and an EC50 of 0.080 nM against human PD-1, as well as an IC50 of 0.75 nM and an EC50 of 0.455 nM against human PD-L1. Reozalimab blocks the binding of PD-1 to PD-L1/PD-L2, as well as the binding of PD-L1 to PD-1/CD80, thereby inhibiting the inhibitory signal transduction of the PD-1 pathway. Reozalimab enhances the functional activation of T cells and induces anti-tumor activity in mouse tumor models reconstituted with human immune cells. Reozalimab can be used in research related to cancers such as non-small cell lung cancer and ovarian cancer.

Reozalimab (1 h) blocks the interactions of PD-1/PD-L2 and PD-L1/CD80, with corresponding IC50 values of 2.27 nM and 0.75 nM, respectively, and also inhibits the binding of PD-1/PD-L1[2].
Reozalimab binds to human and cynomolgus monkey PD-1 and PD-L1 with high affinity, but does not bind to mouse or rat PD-1 or PD-L1[2].
Reozalimab (1 h) specifically binds to CHO-K1 cells expressing PD-1 and PD-L1, with corresponding EC50 values of 2.39 nM and 0.589 nM[2].
Reozalimab (6 h) enhances T cell activation in the co-culture system of CHO-K1 cells expressing PD-L1 or co-expressing PD-L1+PD-L2 with NFAT-luc2/PD1 Jurkat cells, with EC50 values of 0.12 nM and 0.18 nM, respectively[2].
Reozalimab (66 h) enhances the activation of naive human CD4 T cells in allogeneic MLR, with an EC50 of 0.01 nM[2].
Reozalimab enhances primary human pan-T cell-mediated killing of HEL 92.1.7 tumor cells, with an EC50 of 0.005 nM[2].
