Biological Activity
2-Methylbutyryl-CoA (0.1-50 μM) is the substrate for maximal enzymatic activity of purified recombinant potato 2MBCD (Km = 1.0 μM), while human IVD mutants modified to match 2MBCD's active site residues show altered specificity toward 2-methylbutyryl-CoA, with the quintuple mutant losing isovaleryl-CoA activity entirely[1].
2-Methylbutyryl-CoA is accepted as a starter substrate by recombinant LipPks1+TE from Streptomyces aureofaciens Tü117, catalyzing the formation of 3-hydroxy-2,4-dimethylhexanoate with a kcat/KM of 237.0 M-1s-1[2].
