Nampt-IN-22
Nampt-IN-22 is a nicotinamide phosphoribosyltransferase (NAMPT) inhibitor. Nampt-IN-22 blocks the NAD+ salvage synthesis pathway by inhibiting nicotinamide phosphoribosyltransferase (NAMPT). Its tertiary alcohol forms hydrogen bonds with His191 of NAMPT and enhances lysosomal stability via intramolecular hydrogen bonds. As an ADC payload, Nampt-IN-22 exhibits low nanomolar cytotoxicity against tumor cells after conjugation to an anti-CD30 antibody, prolongs survival in a disseminated L540 Hodgkin lymphoma mouse model, and suppresses subcutaneous tumor growth. Nampt-IN-22 can be used in studies related to Hodgkin lymphoma.

Nampt-IN-22 (compound 3) (24 h) exhibits partial resistance to acid-induced hydration into inactive guanylurea metabolites, with only 23% conversion observed after treatment at pH < 2 and 37°C[1].
Nampt-IN-22 (72 h) produces no detectable levels of the inactive guanidino urea metabolite after incubation with human liver lysosomal extracts at 37°C, indicating its resistance to enzyme-driven lysosomal conversion[1].
Free Nampt-IN-22 (0.0001-10 µM; 96 h) inhibits the viability of CD30-positive human Hodgkin's lymphoma L540 cells, with an IC50 of 0.1395 µM, and achieves complete cell killing at high concentrations[1].
