LIBX-A401 is a selective long-chain acyl-CoA synthetase 4 (ACSL4) inhibitor with a human IC50 values of 0.38 μM and a Kd of 0.72 μM. LIBX-A401 binds to ACSL4 in an ATP-dependent manner, stabilizes the C-terminal domain, alters the fatty acid gate region, and interacts with residues A329 and Q302 within the fatty acid binding site. LIBX-A401 exhibits anti-ferroptosis properties in cells. LIBX-A401 can be used for the researches of cancer and parkinson's disease.

LIBX-A401 () (0.38 ± 0.03 μM; up to 50 μM; 10 μM) inhibits wild-type recombinant human ACSL4 with an IC50 of 0.38 ± 0.03 μM and a Kd of 0.72 ± 0.12 μM, while showing no activity against recombinant human ACSL3 (IC50 >50 μM) and no activity against PPARγ at 10 μM[1].
LIBX-A401 inhibits recombinant human ACSL4Q302M, ACSL4Q302A, and ACSL4L325F mutants with IC50 values of 7.1 ± 0.6 μM, 2.4 ± 0.4 μM, and 17.8 ± 3.8 μM, respectively, Kd values of 11.0 ± 1.6 μM, 8.6 ± 1.3 μM, and 43.5 ± 5.4 μM, respectively[1].
LIBX-A401 (5 μM) induces an ATP-dependent thermal shift of wild-type recombinant human ACSL4, increasing its thermal stability by a ΔTm of 5.3 ± 0.2 °C at a concentration of 5 μM[1].
LIBX-A401 (100 μM) specifically binds to the fatty acid gate domain-containing peptide 313DTYIGYLPLAHVLELTAEISCFTYGCR339 of recombinant wild-type human ACSL4 in an ATP-dependent manner, as shown by competitive reduction of photoaffinity probe labeling[1].
LIBX-A401 (2.5 μM; 24 h pre-incubation) protects HEK293 and HT-1080 cells from RSL3 (HY-100218A)-induced ferroptosis[1].
LIBX-A401 (2.5 μM; 4 h pre-incubation) protects LUHMES cells from Arachidonic acid (HY-109590) + Fe-induced ferroptosis and reduces lipid peroxidation[1].
LIBX-A401 (50 μM) does not exhibit DPPH scavenging activity[1].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (7.37 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (7.37 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
