Di-4-ANEPPDHQ is a polarity-sensitive membrane probe with a doubly positive charge that can be used to visualize membrane microdomains in living Arabidopsis cells. Di-4-ANEPPDHQ produces a fluorescence spectrum shift by changing the polarity of the lipid environment after embedding into the cell membrane, thereby quantifying the degree of lipid accumulation. The excitation wavelength of Di-4-ANEPPDHQ is 488 nm, and the emission wavelengths are 560 nm and 650 nm in the liquid ordered and disordered phases, respectively. Di-4-ANEPPDHQ is often used for fluorescence imaging studies of membrane microstructure, lipid order, and membrane tension response in living cells[1][2][3].

NR12S is a fluorescent probe that specifically binds to the outer leaflet of membranes. NR12S exhibits phase-sensitive red edge excitation shift, emission wavelength shift and lifetime changes, which depend on membrane phase, cholesterol content and lipid acyl chain properties. NR12S can be used to study cholesterol and lipid order in biological membranes .

DOPE-Mal is an N-maleimide-derived phosphatidylethanolamine. DOPE-Mal does not interfere with fatty acyl chains or alter lipid bilayer permeability; it increases lipid bilayer order, reduces fluidity, induces conformational changes in phospholipid polar heads and enhances their hydration. DOPE-Mal can be used as a component to prepare maleimide-displaying liposomes, enabling peptide conjugation via maleimide-thiol coupling. DOPE-Mal can be used to prepare Liposomes for delivering Doxorubicin (HY-15142A) to hepatocytes. DOPE-Mal is applicable to research related to hepatocellular carcinoma and lung cancer .

NRERcl is a Golgi-targeting lipid order probe. NRERcl can be used for imaging the lipid order and polarity of the endoplasmic reticulum. NRERcl carries a propyl chloride targeting group that directs it to the endoplasmic reticulum, and it functions via excited-state charge transfer, emitting colors according to local polarity changes associated with lipid order. NRERcl partitions into lipid membranes, with a higher fluorescence quantum yield in membrane environments than in aqueous buffers; its excitation wavelength is 520 nm, and the emission maximum varies with the environment (Ex/Em = 520/584-653 nm) .

NRLyso is a lysosome-targeted solvatochromic fluorescent probe derived from Nile Red (HY-D0718). It can localize to lysosomes and is applicable for imaging lysosomal lipid order and polarity. The excitation wavelength of NRLyso is 520 nm, and its emission maximum varies with the environment. NRLyso partitions well into lipid membranes: it exhibits fluorescence enhancement and emission blue-shift in low-polarity, tightly packed lipid environments, while shows emission red-shift in more polar, disordered lipid environments. NRLyso maintains organelle specificity under oxidative and hyperosmotic stress conditions, and can detect changes in lysosomal lipid order and polarity in response to these stresses, cholesterol extraction, and cholesterol enrichment .

NRERf is an endoplasmic reticulum (ER)-targeted solvatochromic fluorescent probe based on Nile Red (HY-D0718). It possesses ER-targeting capability and can sense changes in local polarity and lipid order of biological membranes. NRERf undergoes excited-state charge transfer, with its emission color changing according to local polarity: the tightly packed, low-polarity liquid-ordered phase (Lo) induces a blue-shifted emission, while the high-polarity liquid-disordered phase (Ld) induces a red-shifted emission. The emission wavelength of NRERf varies with the environment. NRERf can be used to detect changes in lipid order and polarity induced in the ER by cholesterol extraction/enrichment, oxidative stress, and hyperosmotic shock .
