Leupeptin acid is reduced to Leupeptin (HY-18234) by solubilized leupeptin acid reductase from Streptomyces roseus MA839-A1 in an ATP- and NADPH-dependent manner; the enzyme has K_m values of 4.2 × 10^-5 M for ATP and 1.3 × 10^-6 M for NADPH, and is strongly suppressed by leupeptin and antipain rather than elastatinal^[2].
Leupeptin acid is biosynthesized as an intermediate to form leupeptin by the combined leucine acyltransferase and leupeptin acid synthetase-reductase complex from Streptomyces roseus MA839-A1 using acetyl-CoA, L-leucine (HY-N0486) and L-arginine (HY-N0455) as substrates^[2].
