5(S)-HPETE is a 5-lipoxygenase substrate and Arachidonic acid (HY-109590) metabolite. 5(S)-HPETE forms via 5-LOX-catalyzed hydroperoxidation of arachidonic acid. 5(S)-HPETE undergoes 5-LOX-catalyzed epoxidation (dehydration) to form leukotriene A4. 5(S)-HPETE can be used for the research of asthma, rheumatoid arthritis.

5(S)-HpETE (1-30 μM) acts as a substrate for recombinant human 5-LOX-mediated epoxidation, with a Km of 14 μM in the absence of ATP, and ATP induces 4.8-fold activation of Vmax and 3.9-fold activation of Vmax/Km for this reaction[1].
5(S)-HPETE (0.5-40 μM) acts as a substrate for human h12-LOX, with a kcat/KM of 0.13 s−1 μM−1[2].
5(S)-HPETE (0.5-40 μM) acts as a substrate for human h15-LOX-1, with a kcat/KM of 0.23 s−1 μM−1, and the enzyme exhibits altered positional specificity to produce mostly 5S,12S-diHpETE[2].
