S34-1035 is a selective AKR1C3 inhibitor with an IC50 of 0.04 μM. As a chemosensitizer, S34-1035 restores the sensitivity of doxorubicin-resistant breast cancer cells to doxorubicin. S34-1035 can be used in breast cancer research.

Biological Activity
S34-1035 potently and selectively inhibits purified recombinant AKR1C3 with an IC50 of 0.04 μM, exhibiting over 2500-fold selectivity over AKR1C1, AKR1C2, and AKR1C4[1].
S34-1035 (0-100 μM; 72-96 h) exhibits weak antiproliferative activity against MCF-7/DOX cells alone, with the cell survival rate remaining above 70% after treatment with 100 μM for 96 h[1].
S34-1035 (10-50 μM; 24 h pretreatment, 72 h co-treatment) exerts strong synergistic effects with Doxorubicin (HY-15142A) to reduce the viability of MCF-7/DOX cells, with a combination index (CI) value ranging from 0.145 to 0.637, while it shows no single-agent cytotoxicity at the tested concentrations[1].
Combination treatment with S34-1035 (10 μM; 14 days) and Doxorubicin completely inhibits the colony-forming ability of MCF-7/DOX cells, whereas S34-1035 alone has no effect on colony formation[1].
