Tyk2-IN-26 (compound 18) is a TYK2 inhibitor that suppresses TYK2/STAT signaling induced by IL-23 and IFNα, with IC50 values all below 30 nM. Tyk2-IN-26 is applicable to research related to TYK2, IL-23, type I IFN signaling, as well as inflammatory immunity.

Pretreatment with Tyk2-IN-26 (compound 18) for 60 min inhibits IL-23 (10 ng/mL; 22-24 h)-induced STAT3-dependent SEAP reporter gene activity, with an IC50 of <30 nM[1].
Tyk2-IN-26 exhibits only weak or no significant inhibitory effect on SEAP reporter gene responses induced by IL-2 (4 ng/mL; 24 h) and IFNγ (50 ng/mL; 24 h), with IC50 values >1000 nM for both pathways[1].
Tyk2-IN-26 exhibits an IC50 of >1000 nM against recombinant TYK2-JH1, JAK1-JH1 and JAK2-JH1 in HTRF kinase assays, while its IC50 against JAK3-JH1 ranges from 30 to 300 nM[1].
