Research Areas:

Inflammation/Immunology Cancer

NRX-0492 is an orally active BTK PROTAC degrader, with a binding IC50 of 1.2 nM for both wild-type BTK and BTK T474I, and 2.7 nM for BTK C481S, and exhibits cellular DC50 values of 0.1 nM and 0.2 nM against wild-type BTK and BTK C481S, respectively. NRX-0492 catalyzes the ubiquitination and proteasomal degradation of wild-type and drug-resistant mutant BTK by recruiting the CRBN E3 ubiquitin ligase complex. NRX-0492 inhibits the BCR signaling pathway and its downstream NF-κB/MYC transcriptional program, achieves rapid and sustained degradation in primary CLL cells, and exhibits extremely low cytotoxicity. NRX-0492 degrades BTK, inhibits tumor cell proliferation and activation, and significantly suppresses tumor growth in xenograft models. NRX-0492 can be used in research related to chronic lymphocytic leukemia and diffuse large B-cell lymphoma .

Peptide A5K (INF7-A5K-TAT) is an amphiphilic peptide derived from the HA2-TAT fusion scaffold. Peptide A5K can non-covalently bind to CRISPR ribonucleoproteins and efficiently deliver them to cells, such as primary human T cells, B cells, and NK cells. Peptide A5K enables low-toxicity, precise, and multiplex genome editing, holding great application potential in the field of cell therapy .

Ianalumab (VAY-736) (FUT8-KO) is an anti-BAFF-R monoclonal antibody expressed in CHO cells with the fucosyltransferase 8 gene (FUT8) knocked out. Fucose depletion enhances its B cell clearance capacity. Ianalumab (FUT8-KO) competitively blocks the binding of BAFF to BAFF-R, inhibits the BAFF-mediated alternative NF-κB pro-survival signaling pathway, and abrogates the apoptotic (apoptosis) protective effect of BAFF on cancer cells. Ianalumab (FUT8-KO) can be used in research related to primary Sjögren's syndrome and chronic lymphocytic leukemia .

GW7845 is an orally active non-thiazolidinedione, tyrosine-derived PPARγ agonist. GW7845 is effective at inhibiting voltage-dependent calcium channels (VDCC) and relaxing pressurized arteries with IC50 of 3 μM by using Ba 2+ as the charge carrier through VDCC. GW7845-induced apoptosis is mitochondria- and apoptosome-dependent. GW7845 induces rapid mitochondrial membrane depolarization and release of cytochrome c in primary pro-B cells and BU-11 cells .

E234G HYPE-IN-1 (Compound I2.10) is an inhibitor of AMPylation directed by the Huntingtin-interacting protein E (HYPE), possessing low micromolar bioactivity, low cytotoxicity, and blood-brain barrier permeability. E234G HYPE-IN-1 can inhibit the AMPylation of B-cell immunoglobulin-binding protein (BiP), the primary substrate of HYPE. E234G HYPE-IN-1 is suitable for development as a HYPE-specific therapeutic agent for clinical conditions such as neurodegenerative diseases and diabetes .

DXL625 is an autophilic CD20-targeting agent. DXL625 triggers downstream apoptosis signaling pathways dependent on intact lipid rafts and extracellular Ca 2+. DXL625 induces caspase-mediated apoptosis in cancer cells and selectively targets cells in the actively proliferative S phase. DXL625 mediates complement-dependent cytotoxicity in cancer cells and primary B lymphocytes. DXL625 can be used in research related to Burkitt's lymphoma and B-cell lymphoma .

SRX3305 is an BTK/PI3K/BRD4 inhibitor with IC50s of 6.5 nM, 15 nM, and 4 nM toward BTK, PI3Kɑ and PI3Kδ, respectively. SRX3305 attenuates chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL) cell proliferation and promotes apoptosis in a dose-dependent fashion. SRX3305 yields potent anti-tumor effects but spares healthy bystander cells. SRX3305 inhibits the activation-induced proliferation of primary CLL cells in vitro and effectively blocks microenvironment-mediated survival signals. SRX3305 blocks CLL cell migration toward CXCL-12 and CXCL-13. SRX3305 maintains its anti-tumor effects in Ibrutinib (HY-10997)-resistant CLL cells. SRX3305 can be used for research in CLL, diffuse large B-cell lymphoma (DLBCL) and MCL .
