5-Ethynyluridine (5-EU) is a potent cell-permeable nucleoside can be used to label newly synthesized RNA. 5-Ethynyluridine can be used for isolation and sequencing of nascent RNA from neuronal populations in vivo. 5-Ethynyluridine can be used to identify changes in transcription in vivo in nervous system disease models^[1]^[2]. 5-Ethynyluridine is a click chemistry reagent, it contains an Alkyne group and can undergo copper-catalyzed azide-alkyne cycloaddition (CuAAc) with molecules containing Azide groups.

5-Ethynyluridine (75 mM in PBS for 1 μl; intracerebellar injection ) shows a labeling of central nervous system cells in mouse^[1].
5-Ethynyluridine (330-750 μMol/g; i.p.) results in in labelling of nascent RNA in cells of multiple organs and tissues, but not in the central nervous system in mouse^[1].

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 0.71 mg/mL (2.65 mM); Clear solution

This protocol yields a clear solution of ≥ 0.71 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (7.1 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Protocol 2

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

Solubility: ≥ 0.71 mg/mL (2.65 mM); Clear solution

This protocol yields a clear solution of ≥ 0.71 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (7.1 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Protocol 3
