Tris(3-hydroxypropyltriazolylmethyl)amine (THPTA) is an accelerating ligand in the copper-catalyzed azide-alkyne cycloaddition reaction (CuAAC) and protects cells from oxidants generated by copper-catalyzed reduction of oxygen by ascorbate. In addition, Tris(3-hydroxypropyltriazolylmethyl)amine can also protect the histidine moiety of biomolecules in a manner proportional to the ligand concentration.

Biological Activity
Tris(3-hydroxypropyltriazolylmethyl)amine (1.25 mM, 1 h) intercepts and quickly reduce reactive oxygen species generated by the ascorbate-driven reduction of dissolved O2 without compromising the CuAAC reaction rate very much[1].
Tris(3-hydroxypropyltriazolylmethyl)amine (250 μM, 1-5 min) reverses the toxic effect of Cu-ascorbate effect in a dose-dependent manner at a 5:1 ligand:Cu ratio, preserving the viability of HeLa, Chinese hamster ovary (CHO), and Jurkat cells[2].
Tris(3-hydroxypropyltriazolylmethyl)amine (125 μM, 5 min) accelerates the click reaction and allows the cells to survive at a copper concentration of 50 μM in HeLa cells[4].
