8-(4-Chlorophenylthio)-cAMP
8-(4-Chlorophenylthio)-cAMP (8CPT-cAMP) is a cell membrane-permeable cAMP analog with excellent membrane permeability, which activates Sgk kinase via phosphorylation mediated by PKA. 8-(4-Chlorophenylthio)-cAMP is hydrolyzed by intracellular enzymes to produce the active metabolite 8CPT-Ade, which induces cortisol synthesis and the expression of steroidogenic protein genes. When used in combination with insulin, 8-(4-Chlorophenylthio)-cAMP enhances the activation of Sgk; it activates the cAMP pathway to promote the proliferation and iodine uptake of thyroid cells. 8-(4-Chlorophenylthio)-cAMP can be used in studies related to metabolism and signal transduction.

8-(4-Chlorophenylthio)-cAMP (8CPT-cAMP) (0.2 mM; 60 min) activates the activity of wild-type Myc-Sgk to 2-fold in COS7 cells via a PKA-dependent mechanism requiring PDK-mediated phosphorylation; it fails to activate the Thr369→Ala mutant Myc-Sgk and Ser422→Ala mutant Myc-Sgk, indicating that Thr369 and Ser422 are essential for cAMP-mediated activation; it further enhances the activity of wild-type Myc-Sgk activated by Okadaic acid (HY-N6785) in COS7 cells[1].
8-(4-Chlorophenylthio)-cAMP (0.2 mM; 60 min) increases the intensity of the fastest-migrating Myc-Sgk band (B1) in COS7 cells expressing wild-type, Thr369→Ala mutant, or Ser422→Ala mutant Myc-Sgk in a PKA- and PDK-independent manner[1].
8-(4-Chlorophenylthio)-cAMP (0.2 mM; 60 min) induces hyperphosphorylation of wild-type Myc-Sgk in COS7 cells, forming a slower-migrating B3 band that correlates with enzyme activation[1].
8-(4-Chlorophenylthio)-cAMP (0.1 mmol/L; 4 days) stimulates the proliferation of FRTL-5 rat thyroid cells, and this effect is attenuated by 1,25-(OH)2D3 in a dose-dependent manner[2].
8-(4-Chlorophenylthio)-cAMP (0.1 mmol/L; 2-4 days) stimulates iodine uptake by FRTL-5 rat thyroid cells, and this effect is attenuated by 1,25-(OH)2D3 in a dose-dependent manner[2].
8-(4-Chlorophenylthio)-cAMP (increasing concentrations; 3 days) stimulates iodine uptake in FRTL-5 rat thyroid cells in a dose-dependent manner in vitro, whereas 10 nmol/L 1,25-(OH)2D3 attenuates this uptake effect[2].
