WRN-IN-25

WRN-IN-25 is an allosteric Werner syndrome helicase (WRN) inhibitor with an IC₅₀ of 15 nM and a K_d of 54 nM. WRN-IN-25 induces DNA damage, reduces cell viability, and exhibits synthetic lethality in WRN-driven high microsatellite instability cancer cells. WRN-IN-25 (Compound (AC2) (20 μmol/L top concentration, 1:2 serial dilutions; 72 h) shows that HCT116 cells with WRN^I852F mutations are resistant to WRN-IN-25, with WRN-IN-25 inhibiting mutant WRN helicase-mediated DNA unwinding with an IC₅₀ of 312 nM^[1].
