2-Thiocytidine is a natural thionucleoside in prokaryotic tRNA, which is biosynthesized under the catalysis of intracellular sulfur transferase systems and possesses multiple activities such as metal coordination and nucleic acid conformation regulation. 2-Thiocytidine forms stable Zn2+ and Cd2+ complexes via its (C2) S group, with partial involvement of N3; metal binding acidifies the deprotonation process of (C4) NH2. 2-Thiocytidine can be used in related research in various fields including nanotechnology and click chemistry for interstrand DNA crosslinking.

2-Thiocytidine (C2S) has a pKa value of 3.44 in aqueous solution and exhibits stronger acidity than monoprotonated cytidine; deprotonation of this molecule occurs in water with a corresponding pKa of 12.65[1].
2-Thiocytidine (0.4 mM) forms Zn (C2S)2+ and Zn (C2S-H)+ complexes in aqueous solution, where the (C2) S serves as the primary binding site for Zn2+[1].
2-Thiocytidine (0.4 mM) forms Cd (C2S)2+ and Cd (C2S-H)+ complexes in aqueous solution, where (C2) S serves as the primary binding site for Cd2+[1].
2-Thiocytidine can be efficiently incorporated into a DY647-labeled, triple-modified 17-mer RNA sequence via solid-phase phosphoramidite synthesis[2].
