2,3-Butanediol is microbially derived compound that is a discriminating urinary biomarker of Fmo5-/- mice. 2,3-Butanediol prevents age-related increases in the plasma concentration of cholesterol. 2,3-Butanediol has potential applications in the manufacture of printing inks, perfumes, fumigants, moistening and softening agents, plasticizers, and as a carrier for pharmaceuticals.

2,3-Butanediol contains two stereo centers, resulting in three stereo isomers including (2R,3R)-, meso- and (2S,3S)-forms[1].

Short- (4-week) and long-term (14-week) treatment of wild-type mice with a 2,3-Butanediol isomer mix decrease plasma cholesterol and epididymal fat deposition but have no effect on plasma concentrations of glucose or insulin, or on body weight. In the case of long-term treatment, the effects are maintained after withdrawal of 2,3-Butanediol. Short-, but not long-term treatment, also decreases plasma concentrations of triglycerides and non-esterified fatty acids[2].
The isomers of 2,3-Butanediol are discriminating urinary biomarkers of Fmo5-/- mice. Antibiotic treatment of Fmo5-/- mice increased plasma cholesterol concentrations and substantially reduced urinary excretion of 2,3-Butanediol, showing that gut bacteria contribute to the lower plasma cholesterol of Fmo5−/− mice, and that 2,3-Butanediol is microbially derived[2].

Dosage:60 mg/kg/d, 250 mg/kg/d or 600 mg/kg/d (4:1:1 ratio of meso-2,3-Butanediol : 2R,3R-Butanediol : 2S,3S-Butanediol)

Administration:Orally in drinking water; for 4 weeks

Result:At a dose of 250 mg/kg/d, plasma concentrations of both total cholesterol and HDL cholesterol were significantly lower than those of mice that were untreated for 4 weeks.
At doses of 60 or 600 mg/kg/d did not prevent age-related increases in plasma concentrations of either total or HDL cholesterol.

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (27.74 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: 2.5 mg/mL (27.74 mM); Suspended solution; Need ultrasonic
This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
