24-Methylenecholesterol (Ostreasterol) is a regulator targeting acyl-CoA cholesterol acyltransferase (ACAT) with anti-aging and neuroprotective effects. 24-Methylenecholesterol mimics the effects of nerve growth factor (NGF), can extend yeast lifespan through an anti-oxidative stress mechanism, and exhibits neuroprotective activity in PC12 cells. 24-Methylenecholesterol can reduce intracellular reactive oxygen species (ROS) and malondialdehyde (MDA) levels, activate anti-oxidative stress pathways (such as UTH1, SOD-related genes), and promote synaptic growth.

In Vivo

24-Methylenecholesterol (0.1-1.0% dry feed concentration; dietary supplementation; once daily; 3 weeks) significantly upregulates the expression of head nutritional marker proteins (such as MRJP1, MRJP4-7), improves antioxidant enzyme activities and promoted larval development in the Apis mellifera model^[2].

Animal Model:

Apis mellifera worker bees (female, newly emerged, 80-100 mg body weight), nutritional physiology model^[2]
Dosage:

0.1% (S1), 0.25% (S2), 0.5% (S3), 0.75% (S4), 1.0% (S5) 24-Methylenecholesterol dissolved in acetone and mixed into synthetic diets (final acetone concentration <0.5%)
Administration:

Oral administration via synthetic diet, replaced daily, treatment for 3 weeks starting from newly emerged adulthood
Result:

Treatment with 24-Methylenecholesterol at 0.1-1.0% dry diet concentration significantly altered the head proteome of worker bees.
Compared to the control group, the mean relative abundances of major royal jelly proteins (MRJP1, MRJP4, MRJP5, MRJP7)-key nutritional markers involved in brood food synthesis-were upregulated by 20-35% in the S3-S5 groups (0.5-1.0% sterol).
