4-Methoxybiphenyl is a cytochrome P450-dependent microsomal monooxygenase system substrate and metabolite precursor. 4-Methoxybiphenyl undergoes O-demethylation to generate 4-hydroxybiphenyl. 4-Methoxybiphenyl undergoes sulphation and glucuronidation conjugation; inhibition of sulphation shifts metabolism toward increased glucuronidation and unconjugated 4-hydroxybiphenyl accumulation. 4-Methoxybiphenyl serves as a model substrate for studying phase II metabolic conjugation pathway balance in rat isolated hepatocytes.

4-Methoxybiphenyl (100 μM; 20 min) metabolism by isolated male Wistar rat hepatocytes produces 4-OHBP via O-demethylation, which is conjugated into sulphate and glucuronide conjugates; inhibition of sulphation (via 50 μM DCNP or sulphate-free medium) significantly increases 4-OHBP glucuronide, unconjugated 4-OHBP, and total 4-OHBP levels, with total 4-OHBP reaching 19.47 nmol/2 × 106 cells/20 min with 50 μM DCNP and 20.01 nmol/2 × 106 cells/20 min in sulphate-free medium[1].
