Notably, ZRANB1 acts as a positive regulator of the Wnt signaling pathway by deubiquitinating APC protein, a negative regulator of Wnt-mediated transcription. Moreover, it plays a pivotal role in autophagy regulation through the deubiquitination of PIK3C3/VPS34, facilitating autophagosome maturation. Beyond its involvement in intracellular processes, ZRANB1 is implicated in the modulation of cell morphology and cytoskeletal organization, contributing to stress fiber dynamics and cell migration.
