RIBOTAC c-Myc Apoptosis

Research Areas:

MYC-RIBOTAC is a nucleic acid-targeting degrader (ribonuclease-targeting chimera, RIBOTAC) that targets the MYC internal ribosome entry site (IRES). MYC-RIBOTAC contains a MYC mRNA binding component and a small molecule that recruits and locally activates RNAse L1. MYC-RIBOTAC reduces MYC mRNA and protein expression levels, induces cell apoptosis, and can be used for antitumor research . MYC-RIBOTAC consists of pre-miR-155 binder Anticancer agent 167 (HY-156839), RNA binder NCI-B16 (HY-156215), and Linker Amino-PEG4-alcohol (HY-W008005).

BTYNB is a structure-specific nucleic acid binder and IGF2BP1 inhibitor (with an IC50 of 5 μM against hBTYNB). BTYNB disrupts the IGF2BP1-RNA interaction and blocks its binding to oncogenic mRNAs such as c-Myc, MDM2, PD-L1. BTYNB completely blocks the INHBA-Smad2/3 pathway, disrupts the MYCN/IGF2BP1 loop, and thereby induces apoptosis and cell cycle arrest, effectively inhibiting the proliferation and survival of cancer cells. In addition, BTYNB acts as an immune activator and tumor microenvironment modulator, enhances T cell-mediated tumor killing, and produces significant synergistic effects with inhibitors of PD-1, BRD and BIRC5. BTYNB can be used in relevant research on various malignant tumors including ovarian cancer, neuroblastoma, leukemia and melanoma .

2-5A (2′,5′-ApApA) pentasodium solution 100 mM is a high-affinity (Ka=0.04 nM) RNase L binder. 2-5A pentasodium solution 100 mM activates the nuclease activity of latent RNase L to cleave single-stranded RNA by inducing conformational changes and dimerization of latent RNase L, thereby mediating critical antiviral responses and enhancing interferon effects. 2-5A pentasodium solution 100 mM not only effectively inhibits Rauscher murine leukemia virus replication, reduces viremia and splenomegaly, but also enhances antileukemic efficacy when combined with amphotericin B and exhibits favorable high-dose tolerance. 2-5A pentasodium solution 100 mM is an important tool molecule for investigating the pathological mechanisms of viral infection, hereditary prostate cancer and leukemia .

rTRD01 is an orally active, specific TDP-43 binder that targets the RRM1 and RRM2 domains of TDP-43. rTRD01 partially disrupts the interaction between TDP-43 and c9orf72 repeat RNA, but does not affect the binding of TDP-43 to canonical binding sequences. rTRD01 exhibits significant neuroprotective effects in zebrafish models, improves motor function and protects against paraquat (a widely used herbicide)-induced neurodegeneration, with no teratogenicity at high concentrations. rTRD01 is not a general antioxidant and cannot counteract Rotenone (HY-B1756)-induced neuronal death. rTRD01 can be used to study amyotrophic lateral sclerosis and other TDP-43 proteinopathies .

RNA binder 1 (Compound 4b) is a blood-brain permeable RNA binder. RNA binder 1 can selectively bind to the G-quadruplex structure of the G4C2 repeat sequence RNA of the C9orf72 gene. RNA binder 1 significantly reduces the levels of toxic polypeptides poly(GA) and poly(GP) produced by the G4C2 repeat sequence in amyotrophic lateral sclerosis (ALS) patient-derived cells. RNA binder 1 has no significant effect on the antisense polypeptide poly(PR), showing selectivity for sense RNA. RNA binder 1 can be used in the study of ALS and frontotemporal dementia (FTD) .

Guanidino-G-Clamp-PNA is a highly efficient sequence-specific RNA binder and gene silencer. Guanidino-G-Clamp-PNA precisely targets such targets as miR-155 or transthyretin (TTR) mRNA through base pairing: the former regulates tumor-related signaling pathways by reducing microRNA activity, while the latter inhibits the translation of harmful proteins via steric hindrance. Guanidino-G-Clamp-PNA effectively stabilizes DNA/RNA duplexes, induces cancer cell apoptosis, and suppresses tumor growth. In addition, Guanidino-G-Clamp-PNA can be conjugated with targeting ligands to improve tissue-specific delivery and reduce in vivo adverse reactions, and it can also enhance the splicing regulation efficacy of other oligonucleotide platforms (such as PMO) when integrated into them. Guanidino-G-Clamp-PNA is applicable to the research of various diseases including diffuse large B-cell lymphoma and hereditary transthyretin-related amyloidosis .
