  3. Antiviral agent 86

Antiviral agent 86 is an anti-coronavirus agent. Antiviral agent 86 acts as a binder of coronavirus non-structural protein 15 (nsp15), with a Ka value of 67 μM against human targets. Antiviral agent 86 inhibits the replication of HCoV-229E and SARS-CoV-2. Antiviral agent 86 exerts inhibitory effects at the post-entry lifecycle stage of coronaviruses in host cells and inhibits the formation of viral double-stranded RNA (dsRNA). Antiviral agent 86 exhibits an additive antiviral effect when used in combination with GS-441524. Antiviral agent 86 can be used in studies related to coronavirus infections.

Description

Antiviral agent 86 can be used in studies related to coronavirus infections[1].

In Vitro

Antiviral agent 86 (Compound 2p) (5 days) potently inhibits the replication of HCoV-229E in HEL299 cells, with an EC50 of 9.3 μM, and no cytotoxicity is detected at a concentration of 100 μM[1].
Antiviral agent 86 (5 days) inhibits the replication of SARS-CoV-2 in A549ACE2/TMPRSS2 cells, with an EC50 of 12 μM, and no cytotoxicity is detected at a concentration of 100 μM[1].
Antiviral agent 86 (1-5 days) exhibits pan-coronavirus activity and acts on human and animal coronaviruses belonging to the α and β genera, with EC90 values ranging from 2.2 μM (HCoV-OC43 in HRT-18G cells) to 30 μM (TGEV in ST cells). No cytotoxicity is detected at a concentration of 100 μM across all tested cell lines[1].
Antiviral agent 86 (20 μM, 1 day) potently inhibits the production of viral dsRNA in HCoV-229E-infected 16HBE cells, indicating that it is capable of suppressing viral replication[1].
Antiviral agent 86 (0.205-50 μM; 5 days) exhibits additive antiviral activity when used in combination with GS-441524 (HY-103586) in HCoV-229E-infected HEL299 cells[1].
Antiviral agent 86 (5 days) shows reduced potency against nsp15 mutant (V42A or F230L) HCoV-229E in HEL299 cells, with its EC90 values increasing to 7.7 μM and 9.4 μM, respectively[1].
Antiviral agent 86 (6.25-200 μM; 120-second association, 360-second dissociation) binds directly to recombinant HCoV-229E nsp15 protein, with a KD value of approximately 67 μM[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Real Time qPCR[1]

Cell Line: HEL299 cells (HCoV-229E), Vero118 cells (HCoV-NL63), HRT-18G cells (HCoV-OC43), A549ACE2/TMPRSS2 cells (SARS-CoV-2 20A.EU2, SARS-CoV-2 Omicron BF.7), L2 cells (MHV-A59), ST cells (TGEV)
Concentration: serial dilutions; 100 μM (cytotoxicity test)
Incubation Time: 1-5 days (depending on virus)
Result: Demonstrated pan-coronavirus activity with EC90 values of 5.3 μM (infectious titer) and 6.7 μM (RT-qPCR) for HCoV-229E, 6.8 μM for HCoV-NL63, 2.2 μM for HCoV-OC43, 10 μM for SARS-CoV-2 20A.EU2, 14 μM for SARS-CoV-2 Omicron BF.7, 5.5 μM for MHV-A59, and 30 μM for TGEV.
Showed no cytotoxicity at 100 μM in any of the tested cell lines.

Immunofluorescence[1]

Cell Line: human bronchial epithelial 16HBE cells infected with HCoV-229E
Concentration: 20 μM
Incubation Time: 1 day post-infection
Result: Strongly suppressed the dsRNA signal in infected cells, comparable to the effect of the reference polymerase inhibitor GS-441524.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
