  • Function

    The LRP4 protein, a member of the low-density lipoprotein receptor-related protein family, plays a critical role in multiple developmental and physiological processes. It functions as a co-receptor in the Wnt signaling pathway, particularly in the canonical Wnt/β-catenin cascade, where it facilitates the interaction between Wnt ligands and Frizzled receptors, thereby modulating downstream gene expression[1]. LRP4 is essential for neuromuscular junction (NMJ) formation and maintenance, acting as a key regulator in presynaptic differentiation and postsynaptic clustering of acetylcholine receptors[2]. In addition to its role in neural development, LRP4 has been implicated in bone metabolism, where it regulates osteoblast differentiation and mineralization through interactions with sclerostin and other bone-related factors[3]. Recent studies also highlight its involvement in cardiovascular function, including endothelial cell integrity and vascular smooth muscle cell behavior. Furthermore, LRP4 has emerged as a potential therapeutic target in autoimmune diseases such as myasthenia gravis, where autoantibodies against LRP4 disrupt NMJ stability[4].
