Paspalicine
Paspalicine is an indole diterpenoid fungal metabolite with no tremorgenic activity in animals. Paspalicine targets the large-conductance calcium-activated potassium (maxi-K) channel, blocks channel activity, and modulates the binding of Charybdotoxin (HY-P0191) to the maxi-K channel via a positive allosteric mechanism that enhances toxin binding. Paspalicine can be used in studies related to ryegrass staggers.

Paspalicine allosterically enhances [125I]ChTX binding to maxi-K channels in bovine aortic smooth muscle sarcolemmal membrane vesicles, with a K1/2 of 100 nM and a maximum 120% increase in specific binding relative to control[2].
Paspalicine (10 nM; measured 3 min post-exposure) potently blocks maxi-K channels in excised inside-out membrane patches from bovine aortic smooth muscle cells, with 10 nM causing 83% current block, and the block is partially reversible after washout[2].
