BCN-PEG6-OH

BCN-PEG6-OH is a neutralizing agent and anticoagulant inhibitor containing strained alkyne. BCN-PEG6-OH undergoes strain-promoted alkyne-azide cycloaddition (SPAAC) click reaction with azido-Warfarin to generate an inactive product that can be rapidly cleared via the kidneys. In mice, BCN-PEG6-OH effectively reduces the anticoagulant activity of azido-Warfarin in a dose-dependent manner, normalizes prothrombin time, and exhibits no inherent anticoagulant or procoagulant effects when administered alone. BCN-PEG6-OH can be used for the research of drug-induced coagulopathy.

BCN-PEG6-OH (compound 9) (26-52 mg/kg; subcutaneous injection; 2 doses total at 24-hour intervals) dose-dependently neutralizes azido-Warfarin-induced anticoagulant activity in mice, is cleared via the kidneys, and restores prothrombin time (PT) to levels close to those of the vehicle control group^[1].

Animal Model:

C57BL6N Mice (male, 8-12 weeks of age, drug-induced coagulopathy model via azido-Warfarin administration)^[1]
Dosage:

26 mg/kg; 52 mg/kg
Administration:

s.c.; 2 doses with 24-hour interval, 5 minutes after each i.p. injection of azido-Warfarin
Result:

Reduced the PT fold change (relative to vehicle) from 3.61 (azido-Warfarin alone) to 2.39.
Reduced the PT fold change from 3.61 to 1.75, a level that was not significantly different from vehicle controls.
Led to in vivo formation of the inactive clicked product 10, with plasma concentrations of free azido-Warfarin dropping below the limit of detection within 60 minutes, and 95.3% of the clicked product recovered in urine indicating rapid renal clearance.
