Cdk5i peptide is a CDK5 inhibitory peptide with high binding affinity for Cdk5/p25 (Kd of 0.17 μM), which can block the interaction between CDK5 and p25 and reduce the kinase activity of CDK5/p25. Cdk5i peptide can be used in the stud y of neurodegenerative diseases.

Cdk5i peptide consists of 12 amino acids (ARAFGIPVRCYS) derived from the CDK5 T loop. This 12-amino-acid sequence is highly conserved in CDK5 across species, yet not present in other Cdk family members[1].
Cdk5i peptide binds to the Cdk5/p25 complex with more than 92-fold higher affinity than to Cdk5 alone (Kd = 0.17 μM vs. Kd = 15.72 μM)[1].
Study using co-expressed Cdk5/p25 and Cdk5i peptides in HEK293T cells. Western blot analysis confirmed that coexpression of Cdk5i peptide does not change the amounts of Cdk5 and p25 protein present in cells. However, Cdk5i peptide coexpression does reduce both the interaction of Cdk5 with p25 (~25%) and Cdk5 kinase activity (~27%), while it did not significantly impact the kinase activity of Cdk5/p35 and Cdk1/Cyclin A complexes[1].
