NSD3/WHSC1L1

NSD3 (WHSC1L1) is a member of the nuclear receptor SET domain (NSD) family of chromatin regulators and functions primarily through histone H3 lysine 36 (H3K36) methylation to control chromatin accessibility and transcriptional programs[1]. Mechanistically, the catalytically active long isoform (NSD3L) promotes deposition of H3K36 methylation marks that support transcriptional activation of genes involved in cell proliferation, differentiation, and oncogenic signaling pathways[1][2]. In disease settings, NSD3 is recurrently amplified or dysregulated across multiple malignancies, and experimental studies have linked NSD3-dependent chromatin remodeling to tumor initiation, progression, and metastatic behavior[1][3]. NSD3 also participates in transcriptional regulatory complexes that coordinate enhancer-associated gene expression, further connecting epigenetic regulation with cancer-associated transcriptional networks[1][2]. Compared with related NSD family members, a key distinguishing feature of NSD3 is the existence of a short isoform (NSD3S) that lacks the catalytic SET methyltransferase domain yet retains important biological activity through protein-protein interactions[1][2]. NSD3S functions as an adaptor that links BRD4 to the CHD8 chromatin remodeler and is required for leukemia maintenance, demonstrating that NSD3 can drive disease through mechanisms independent of catalytic methyltransferase activity[2]. For experimental applications, this isoform-specific biology has motivated development of NSD3-targeted strategies, including compounds directed against functional chromatin-interacting domains, to investigate NSD3-dependent transcriptional and epigenetic processes in cancer models[1].
