DOPE-PEG2000-MG1 is an amphiphilic lipid-PEG-peptide conjugate comprising DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the MG1 peptide. MG1 peptide functions as a tumor-targeting ligand that specifically recognizes proteins overexpressed on tumor cell surfaces. DOPE-PEG2000-MG1 is used to modify liposomes and lipid nanoparticles for active tumor-targeted drug delivery.

DOPE-PEG2000-MG1: surface decoration of liposomes and nanocarriers for tumor-targeted drug delivery, enabling selective accumulation in tumor tissues and enhanced cellular uptake by cancer cells.
Structural composition:
(1) DOPE; a phospholipid with two oleoyl chains that serves as a stable hydrophobic anchor in lipid bilayers and promotes pH-dependent membrane fusion for endosomal escape;
(2) PEG2000; a hydrophilic polymer chain that forms a protective hydration layer reducing immune recognition and extending blood circulation half-life;
(3) MG1 peptide; a targeting ligand that specifically binds to receptors or proteins overexpressed on tumor cell surfaces, acting as a navigation module to direct nanocarriers to tumor tissues for improved therapeutic efficacy and reduced systemic toxicity.
